2. Experimental Evidence for Sustained Quantum Coherence in Microtubules:
The search for direct experimental evidence of sustained quantum coherence within neuronal microtubules, particularly under conditions mimicking the living cell (in vivo or physiologically relevant) and on timescales relevant to neural processing (milliseconds or longer), has yielded several intriguing findings.
One line of evidence stems from studies investigating the effects of anesthetic agents on consciousness. Research has indicated that anesthetics act on microtubules within neurons, and this interaction correlates with a delay in the onset of unconsciousness caused by anesthetic gases.4 Proponents of quantum consciousness theories interpret this as indirect support for the role of microtubules in consciousness, suggesting that anesthetics interfere with quantum processes occurring within these structures that are necessary for conscious experience.4 For instance, one study demonstrated that a microtubule-binding drug delayed unconsciousness in rats under anesthesia, a result that the researchers argued supports the quantum model of consciousness.9 However, it is important to note that alternative explanations exist, suggesting that the observed changes in neuronal activity and consciousness due to microtubule disruption by anesthetics might be attributable to classical mechanisms related to the structural and transport roles of microtubules within neurons.4
More direct evidence has been sought through the detection of quantum vibrations within microtubules. In 2014, Anirban Bandyopadhyay and colleagues reported the detection of quantum vibrations in microtubules, with their lengths varying up to 50 micrometers.1 These findings were claimed to corroborate the Orch OR theory, which posits that consciousness arises from quantum processing in microtubules.12 The reported frequency ranges of these vibrations spanned megahertz, gigahertz, and even terahertz frequencies.12 However, these findings have also been met with controversy within the scientific community, with challenges raised regarding their replication and interpretation.22
A more recent and potentially compelling piece of evidence comes from a 2024 study by Babcock et al., which reported evidence for mesoscale quantum coherence in microtubules through the observation of ultraviolet superradiance from networks of tryptophan, an aromatic amino acid found within tubulin.11 Superradiance is a cooperative quantum effect where multiple emitters, in this case tryptophan molecules, collectively emit photons at an enhanced rate. The observation of this phenomenon in microtubules, even under thermal equilibrium conditions, suggests the presence of quantum coherence over relatively large scales within a biological system.4 The study reported timescales ranging from hundreds of femtoseconds for the brightest states to tens of seconds for the darkest states within these tryptophan lattices.35 This finding challenges the traditional view that such coherence would rapidly decay in the warm and noisy cellular environment and suggests a potential role for these tryptophan networks in photoprotection and ultrafast information transfer within neurons.4
Furthermore, a 2014 study utilized quantum coherence spectroscopy to measure the quantum coherence timescales in microtubule proteins, reporting results consistent with the predictions of the Orch OR theory.23 However, a subsequent review in 2018 raised concerns regarding the methodology and interpretation of this study, indicating that the evidence was not conclusive proof of microtubule quantum processing.23

3. Theoretical Calculations and the Decoherence Problem:
A significant challenge to the hypothesis of functional quantum effects in neuronal microtubules is the issue of quantum decoherence. In 2000, Max Tegmark published calculations suggesting that any quantum coherent system within the brain would undergo extremely rapid decoherence due to its interaction with the warm and wet cellular environment.24 His calculations for microtubules indicated decoherence timescales on the order of 10<sup>-13</sup> to 10<sup>-20</sup> seconds, which are vastly shorter than the timescales relevant for neural processing (milliseconds or longer).24 Based on these results, Tegmark concluded that the degrees of freedom in the human brain related to cognitive processes should be considered classical rather than quantum systems.44
However, these calculations and conclusions have been critically examined by proponents of quantum consciousness theories. Hagan, Hameroff, and Tuszyński argued that Tegmark's analysis was based on a hybrid model that did not accurately represent the Orch OR proposal, which posits superpositions of tubulin dimer conformational states rather than a soliton in superposition along the microtubule.3 After correcting for these differences and considering factors such as superposition separation, charge versus dipole interactions, and the dielectric constant of the environment, they recalculated the decoherence time to be significantly longer, in the range of 10<sup>-5</sup> to 10<sup>-4</sup> seconds.54 They also proposed that incoherent metabolic energy supplied to the collective dynamics ordering water around microtubules could counter decoherence effects, similar to how lasers maintain coherence at room temperature.54 Furthermore, they suggested that phases of actin gelation might enhance the ordering of water around microtubule bundles, further increasing the decoherence-free zone.54
Other theoretical calculations have yielded a range of decoherence times for microtubules based on different mechanisms. Mavromatos and Nanopoulos estimated decoherence times for dipolar excitations in microtubules to be between 0.1 nanoseconds and 0.1 milliseconds, depending on the assumptions used.58 In contrast, Xiang et al. developed a model considering the interaction between microtubules and plasmon in neurons, estimating a much shorter decoherence time scale of approximately 1 to 100 femtoseconds.64 These varying theoretical perspectives underscore the complexity of the issue and the sensitivity of the results to the chosen model and parameters.
The current scientific consensus regarding the severity of the decoherence problem for hypotheses like Orch OR remains divided.23 The mainstream scientific community generally remains skeptical, primarily due to the seemingly inhospitable "warm, wet, and noisy" environment of the brain for maintaining delicate quantum coherence.23 However, recent experimental findings demonstrating longer coherence times in other biological systems have renewed some interest in the possibility of functional quantum effects within the brain.12
4. Proposed Shielding Mechanisms and Alternative Calculations:
To address the decoherence challenge, several mechanisms have been proposed that could potentially shield microtubules from environmental interactions and prolong quantum coherence within the cellular environment.
One such proposal involves the cyclical encasement of microtubules by actin gels.3 This mechanism suggests alternating phases of isolation (when microtubules are encased in a gel-like state of actin polymerization, potentially suitable for quantum computing) and communication (when the cytoplasm is in a more liquid state, allowing for classical interactions).
Another proposed shielding mechanism involves ordered water molecules within the hollow core and surrounding the microtubules.22 The highly structured nature of water near hydrophilic surfaces like microtubules, sometimes referred to as Exclusion Zones (EZ), could potentially create a protected environment that supports quantum coherence. Interactions between these ordered water molecules and the tubulin dimers forming the microtubule lattice might also play a role in maintaining coherence.22
The Debye layer, a layer of counterions that forms around charged biomolecules like microtubules in the cellular environment, has also been suggested as a potential shield against decoherence.27 This layer could screen thermal fluctuations from the bulk cytoplasm, potentially protecting any delicate quantum states within the microtubule. While this screening effect might also reduce the interaction strength between the microtubule and its environment, it could still provide a net benefit for maintaining coherence.64
In addition to these proposed shielding mechanisms, alternative theoretical calculations, such as those by Hagan, Hameroff, and Tuszyński, have yielded significantly longer decoherence times compared to initial estimates, suggesting that the decoherence problem might not be as severe as originally thought.54 Furthermore, the possibility of topological quantum computation occurring within the microtubule lattice has been explored. Topological qubits are theorized to be inherently more resistant to decoherence due to their reliance on the topology of the system rather than the specific state of individual qubits.3
5. Evidence for Microtubule Quantum Computation:
Beyond the potential for sustained quantum coherence, the question remains whether there is specific evidence demonstrating that microtubules actually perform quantum computations that are functionally linked to neuronal activity or cognitive processes.
The Orchestrated Objective Reduction (Orch OR) theory, proposed by Penrose and Hameroff, provides a detailed framework for how microtubules could function as quantum computers within neurons.1 According to this theory, tubulin dimers within microtubules can exist in a superposition of different states, acting as quantum bits (qubits).1 These qubits are proposed to interact through quantum entanglement, evolving according to the Schrödinger equation during dendritic and somatic integration within neurons.21 The quantum computations are then theorized to terminate via Penrose's objective reduction (OR), a quantum state reduction process linked to quantum gravity, resulting in moments of conscious awareness.1 These OR events are proposed to select specific microtubule states that then influence neuronal firing and ultimately control behavior.1
Theoretical models have also explored the possibility of microtubules acting as quantum Hopfield neural networks 107 and supporting topological quantum computation due to their lattice structure.58 However, direct experimental verification of these quantum computational processes within microtubules in living neurons and their functional link to neuronal activity or cognition remains a significant challenge.3 Measuring the quantum states of potential qubits within microtubules, such as the spins of π electrons, is particularly difficult due to their potential entanglement with the surrounding cellular environment, leading to rapid decoherence.92
6. Distinguishing Functional Quantum Effects in Biological Systems:
To discern whether the quantum phenomena observed or proposed in microtubules are functionally relevant and non-trivial, it is essential to distinguish them from the baseline quantum mechanics that governs all biological matter. Quantum mechanics is fundamental to the formation of chemical bonds and the structure of all molecules within biological systems.1 This is often considered "trivial" in the context of quantum biology.169
Functionally relevant, non-trivial quantum effects in biological systems typically involve phenomena such as long-ranged, long-lived, or multiparticle quantum coherences, the explicit use of quantum entanglement, the relevance of single photons or spins triggering macroscopic events, or effects that enhance biological processes beyond what can be explained by classical physics alone.34 Established examples of such effects in biology include quantum coherence in energy transport during photosynthesis, where energy can explore multiple pathways simultaneously to find the most efficient route 1, magnetoreception in migratory birds, where quantum entanglement in radical pairs is thought to allow them to sense the Earth's magnetic field 34, and quantum tunneling in enzyme catalysis, which can significantly enhance reaction rates.34
When considering the evidence for quantum effects in microtubules, the reported superradiance from tryptophan networks suggests a non-trivial quantum coherence extending over mesoscopic scales.11 The proposed role of these effects in ultrafast information transfer and potential links to consciousness through theories like Orch OR also suggest a functional relevance beyond basic molecular interactions.1 However, further research is needed to definitively establish the extent and nature of these effects and their specific contributions to neuronal function and cognition.
7. Alternative Classical Explanations of Observed Phenomena:
For phenomena sometimes attributed to quantum effects in microtubules, established classical or non-quantum biological explanations often exist, and their empirical support is generally stronger due to decades of research in neuroscience and related fields.
For example, while some interpret the effects of anesthetics on microtubules as evidence for quantum involvement in consciousness, the prevailing view in neuroscience attributes the mechanisms of general anesthesia to classical interactions with neural ion channels, receptors, and the lipid membrane environment.6 Extensive pharmacological studies have identified specific binding sites for various anesthetics on these molecular targets, and the correlation between anesthetic potency and lipid solubility (the Meyer-Overton correlation) has been a cornerstone of anesthetic research for over a century.8 Neuroimaging studies have also provided detailed insights into how anesthetics modulate brain activity at the network level.194 While the precise mechanisms of action for all anesthetics are not yet fully elucidated, the empirical support for classical explanations is substantial.
Similarly, the neural correlates of consciousness (NCC) are actively researched within a classical neuroscience framework. Theories such as the Global Workspace Theory, Integrated Information Theory, and Higher-Order Theories propose that consciousness arises from complex neural network activity and information processing within the brain.1 These theories are supported by a vast body of empirical evidence from neuroimaging, lesion studies, and cognitive experiments, linking specific brain regions and neural activity patterns to conscious experience.94 While these classical approaches face their own challenges, particularly in explaining the subjective nature of consciousness (qualia), they offer empirically grounded frameworks for understanding the neural basis of conscious experience.

While experimental evidence, such as the recent observation of superradiance in tryptophan networks within microtubules, suggests the possibility of sustained quantum coherence under physiological conditions, these findings are still relatively new and require further investigation to fully understand their implications. The theoretical challenge posed by quantum decoherence in the warm and wet brain environment remains significant, although proposed shielding mechanisms and alternative calculations offer potential avenues for resolving this issue.
The Orch OR theory provides a detailed framework for how microtubules could perform quantum computations relevant to brain function and consciousness. However, direct experimental evidence demonstrating that microtubules actually process qubits in a way that is functionally linked to neuronal activity or cognition is still limited. The scientific community generally remains skeptical of quantum consciousness theories, with classical neurobiological explanations for phenomena like anesthetic action and the neural correlates of consciousness currently holding stronger empirical support.
Distinguishing between the baseline quantum mechanics present in all biological matter and functionally relevant, non-trivial quantum effects requires careful consideration of criteria such as long-range coherence, entanglement, and enhancement of biological processes beyond classical limits. While some evidence for quantum effects in microtubules appears to meet these criteria, more research is needed to solidify these claims and to differentiate them from potential classical explanations.
Future research in this interdisciplinary field will likely focus on developing more sophisticated experimental techniques to directly probe quantum states and dynamics within microtubules in vivo, as well as refining theoretical models to better understand the interplay between quantum phenomena and classical biological processes.
